Anthranilate synthetase. Some physical and kinetic properties of the enzyme from Serratia marcescens.

چکیده
Anthranilate synthetase from Serratia marcescens was purified to homogeneity. Criteria for homogeneity were disc gel electrophoresis, sedimentation analysis, and immunodiffusion. During electrofocusing the enzyme was found to have an isoelectric pH of 4.6. Glutamineand ammoniadependent activities superimposed during electrofocusing. Glutamine-dependent activity showed a pH optimum of 7.6, while ammonia-dependent activity was optimal above pH 8.5, indicating that NH8 rather than NH*+ was involved in the reaction mechanism. The enzyme was sensitive to feedback inhibition by very low concentrations of Ltryptophan. The mercurial, j-chloromercuribenzoate, was found to be ineffective in desensitizing the enzyme to tryptophan inhibition. However, glutamine utilization was selectively and reversibly blocked. Chorismate enhanced this inactivation, while glutamine and a mixture of glutamine and chorismate were increasingly effective protective agents. A molecular weight of 140,000 was estimated from sucrose gradient centrifugation (7.6 S). The molecular weight on Sephadex gel filtration was 150,000.

Tryptophan biosynthetic pathway in the Enterobacteriaceae: some physical properties of the enzymes.
Several physical properties of the first four enzymatic activities of the tryptophan pathway were examined using gel filtration and ion exchange chromatography. Five different patterns were noted. Differences in the anthranilate synthetase (AS) and phosphoribosylanthranilate transferase (PRT) defined these patterns. In all the organisms studied phosphoribosylanthranilate isomerase and indolegly...
متن کاملThe structures of anthranilate synthase of Serratia
The crystal structure of anthranilate synthase (AS) from Serratia marcescens, a mesophilic bacterium, has been solved in the presence of its substrates, chorismate and glutamine, and one product, glutamate, at 1.95 Å, and with its bound feedback inhibitor, tryptophan, at 2.4 Å. In comparison with the AS structure from the hyperthermophile Sulfolobus solfataricus, the S. marcescens structure sho...

A reversible noncovalent complex is formed prior to irreve...
متن کاملStudy of anthranilate synthase function by replacement of cysteine 84 using site-directed mutagenesis.
Cysteine 84 was replaced by glycine in Serratia marcescens anthranilate synthase Component II using site-directed mutagenesis of cloned trpG. This replacement abolished the glutamine-dependent anthranilate synthase activity but not the NH3-dependent activity of the enzyme. The mutation provides further evidence for the role of active site cysteine 84 in the glutamine amide transfer function of ...
متن کاملSerratia marcescens B4A Chitinase Product Optimization Using Taguchi Approach
Chitinase production by newly isolated Serratia marcescens B4A was optimized following Taguchi’sarray methods. Twenty-three bacterial isolates were screened from shrimp culture ponds in the South ofIran. A chitinase-producing bacterium was isolated based on it’s ability to utilize chitin as the sole carbonsource. The isolate designated as B4A, was identified as Serratia marces...
